چکیده

The low pathogenic avian influenza (LPAI) caused by H9N2 virus has generated economic losses in the Korean poultry industry since 1999, but field isolates could not be used for killed oil emulsion vaccine directly because of low productivity in embryonated chicken eggs (ECE). Therefore, we established a vaccine strain, KBNP-0028, by passage through ECE with allantoic fluid showing the highest volume and titer. KBNP-0028 possessed 158 N-glycan on hemagglutinin (HA) as its parent strain, but acquired a 16 amino acid deletion in the stalk region of neuraminidase (16-del NA) as a result of balancing with the glycosylated HA. KBNP-0028 was sufficiently productive in ECE to be compared with the A/Puerto Rico/8/34 (PR8) strain and immunogenic enough to induce high antibody titers. The antibodies inhibited hemagglutination of recent field isolates as highly as they inhibited KBNP-0028. Genome analysis revealed that KBNP0028 carried no known mutation relevant to virulence in mammalian. In conclusion, KBNP-0028 is a promising vaccine candidate for prevention of the LPAI in the Republic of Korea.

Optimization of incubation temperature in embryonated chicken eggs inoculated with H9N2 vaccinal subtype of avian influenza virus

There are little information about growth properties of low pathogenic (LP) avian influenza virus (AIV) in embryonated chicken eggs (ECEs) at different incubation temperatures. Knowledge of this information increases the quantity and quality of antigen in vaccine production process. For this purpose, 10-5 dilution of AIV (A/Chicken/Iran/99/H9N2) was inoculated (Intra-allantoic) into ...

Development and Evaluation of Real-Time RT-PCR Test for Quantitative and Qualitative Recognition of Current H9N2 Subtype Avian Influenza Viruses in Iran

Avian influenza H9N2 subtype viruses have had a great impact on Iranian industrial poultry production economy since introduction in the country. To approach Rapid and precise identification of this viruses as control measures in poultry industry, a real time probe base assay was developed to directly detect a specific influenza virus of H9N2 subtype -instead of general detection of Influenza A ...

Molecular evaluation of M2 protein of Iranian avian influenza viruses of H9N2 subtype in order to find mutations of adamantane drug resistance

Background: The H9N2 subtype of influenza A viruses is considered to be widespread in poultry industry. Adamantane is a group of antiviral agents which is effective both in prevention and treatment of influenza A virus infections. These drugs inhibit M2 protein ion channel which has role on viral replication. OBJECTIVES: The main objective of this study is to evaluate M gene of avian influenza ...

Genetic and phylogenetic analysis of the ribonucleoprotein complex genes of H9N2 avian influenza viruses isolated from commercial poultry in Iran

BACKGROUND: The H9N2 subtype of avian influenzaviruses (AIVs) has been isolated in multiple avian species inmany European, Asian, African and American countries. Sincethe first outbreak of H9N2 virus in Iran in 1998, this virus haswidely circulated throughout the country, resulting in majoreconomic losses in chicken flocks. Several amino acids in thevirus ribonucleoprotein (RNP) complex includi...

Full Length Characterization of PA Gene of H9N2 Isolated from Broilers During 1998 to 2009

Background and Aims: Avian Influenza (AI) H9N2 subtype was first reported to infect turkeys in the United States in 1966 and has been panzootic in Europe and Asia. The impact of avian influenza caused by H9N2 viruses in Iran is now significantly more severe than in previous years. Methods: Sequence analysis and phylogenetic study of the complete coding region Polymerase A (PA) gene of H9N2 subt...
